Effect of hypermethylated in cancer 1-downstream-regulated gene <em>HMMR</em> on cell migration and invasion in breast cancer: an <em>in vitro</em> study on breast cancer cell line MDA-MB-231

Abstract

Abstract:Objective:To investigate the regulatory relationship between hypermethylated in cancer 1 (HIC-1) and hyaluronan-mediated motility receptor (HMMR) and to study the effect ofHMMRon migration and invasion of MDA-MB-231 breast cancer cell strains.Methods:MDA-MB-231 cells were transfected with double-strand saRNAHIC-1and double-strand siRNAHMMR, respectively. The expression of HIC-1 and HMMR in MDA-MB-231 cells were detected by RT-PCR and Western blotting. CCK-8 assay was used to detect cell proliferation after siRNA or saRNA transfection. Transwell assay was used to determine the effect of siRNA or saRNA transfection on migration and invasion of MDA-MB-231 breast cancer cells.Results:MDA-MB-231 breast cancer cells were successfully transfected. The up-regulation ofHIC-1inducedHMMRdown-regulation, and significantly inhibited cell migration and invasion of MDA-MB-231 breast cancer cells.HMMRwas down-regulated by siRNAHMMRtransfection leading to inhibition of cell migration and invasion. However, cell proliferation have not been affected in both saRNAHIC-1and saRNAHMMRtransfections.Conclusions:HMMRis regulated byHIC-1and it plays an important role in cell migration and invasion. Inhibition ofHIC-1upregulatesHMMR, thus promotes cell migration and invasion of breast cancer cells.
